The system uses an in-situ microscope and an online Raman spectrometer as the Process Analytical Technology (PAT) tools to perform real-time in-situ monitoring and analysis of cell morphology, cell size, cell density, as well as concentrations of cell nutrients and metabolic products during the cell growth process (five biochemical indicators: glucose, lactate, glutamate, glutamine, and ammonium ions). The in-situ microscope captures cell images in real time to obtain the cell diameter distribution and estimate cell density in real time. Through the online Raman spectrometer, the Raman spectra of the system are acquired in real time, and a multivariate calibration model between spectral data and biochemical indicator concentrations is established to analyze the concentrations of biochemical indicators during the cell culture process in real time.
The result from the in-situ microscope exhibits good agreement with those from a cell counter; the multivariate calibration model built between the Raman spectral data and biochemical indicator concentrations can give satisfactory prediction to the concentrations of biochemical indicators in real time.
